The VADEX technology is a self-adjuvanting protein nanoparticle platform. The building block of VADEX technology is a fusion protein combining an amphipathic helical peptide (AHP) and a superfolder GFP (sfGFP). The precisely designed 19 a.a. AHP serves both as a polymerization module and an adjuvant. This fusion protein can be self-assembled into a thermally-stable protein nanoparticle (PNP) that endures ambient temperature storage up to 40 oC for two months. A target antigen can be expressed on the surface of nanoparticle by either loop insertion (VADEX-PEP) or strand complementation (VADEX-Pro) through sfGFP.  Being self-adjuvanting,  VADEX can truly enables Global Vaccination effort that protects every child from Sahara desert to Amazon rain forest. 

Applications: Target peptide with a size less than 90 amino acids can be inserted through loop insertion into the loop9 of sfGFP to form a fusion protein. The fusion protein will self-assemble into protein nanoparticle after protein expression in bacteria and used as antigen for stimulate long-lasting humoral immune response.

Applications: Target protein of any size can be fused with the strand-11 of sfGFP and be incorporated onto the protein nanoparticle when co-expressed with AH-sfGFP-10 fusion protein in bacteria. The purified protein nanoparticle can be used as an antigen for stimulating the induction of IgG with 100 folds higher affinity than antigen with AddaVax.
